Specificity ErbB-2 (also known as Her2 or neu) is a 185 kDa transmembrane receptor tyrosine kinase from the EGFR family that acts to regulate a variety of biological responses including cell growth, differentiation, and tissue development. Ligand binding to the extracellular domain, or overexpression of the receptor leads to phosphorylation on several tyrosine residues within the cytoplasmic domain, and activation. Overexpression or abnormal activation of ErbB-2 has been found in a variety of tumors including brain, breast, lung and skin cancer. Tyrosine 1248 within the cytoplasmic domain of the receptor is an autophosphorylation site that allows binding of Shc and activation of the Ras . Raf . ERK1&,2 signaling pathway. This antibody reacts with Human ErbB-2. Other species (100% homologous) have not been tested, but are expected to react. ErbB-4 (58% homologous) may also cross-react, but has not been tested. In cell lines where EGFR is highly expressed, this antibody may cross-react with EGFR.

Application Western Blotting (WB)
Application Notes Suitable for use in Western blotting . For Western blotting, we recommend using the antibody at 0.1-1.0 µg/mL. At 0.50 µg/mL, the dilution provides 100 mL working solution, which at 10 mL/blot allows 10 blots to be performed. The optimal antibody concentration should be determined for each specific application. Positive control used: SK-BR-3 cell line (overexpresses ErbB-2). Other applications not tested. 1 Peptide Competition: Extracts prepared from SK-BR-3 cells were resolved by SDS-PAGE on a 10% Tris-glycine gel and transferred to PVDF.
