Spezifität The immediated early gene product c-Fos is expressed following mitogenic stimulation. c-Fos functions as a sensor for MAPK signal duration. When MAPK activation is transient, MAPK activity declines before accumulation of the c-Fos protein. When MAPK activation is sustained, c-Fos is phosphorylated by MAPK at serine 374. Phosphorylation stabilizes the Fos protein and primes c-Fos for additional phosphorylation at threonine 325. This antibody specifically interacts with c-Fos phosphorylated at serine 374 and does not crossreact with the non-phosphorylated c-Fos nor with unrelated serine-phosphorylated proteins. The antibody is suitable for Western blot and ELISA applications. Epitope: S L S pS P T L (phosphoserine 374). Reacts with human and dog.

Applikationshinweise Western Blot: 0.5 µg/ml for HRPO/ECL detection Recommended blocking buffer CPPT: 0.5% (w/v) casein, 1% (w/v) PEG 4000, 1% (w/v) Polyvinylpyrrolidone (PVP), 0.1% (v/v) Tween 20, 2x PBS. ELISA: 0.1 µg/ml (protein ELISA), capture ELISA: not determined. Positive control: HepG2 EGF-treated. Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user.
Buffer This purified antibody lyophilized from 1 ml 2 x PBS / 0.09 % Na-azide / PEG and Sucrose. Reconstitute with 1 ml H2O (15 min, RT).
Lagerung For long-term storage, freeze lyophilizate upon arrival (-20°C). Upon reconstitution, aliquote and freeze in liquid nitrogen, reconstituted antibody can be stored frozen at -80°C up to 1 year. Thaw aliquots at 37°C. Thawed aliquots may be stored at 4°C up to 3 months. Avoid repeated freeze-thaw cycles. Shelf life: one year from despatch.
