Background

Green fluorescent protein (GFP) is a protein of natural origin that emits light of a specific wavelength in the lower green portion of the visible light if excited by ultraviolet or blue light. There are different types of GFP as well as modifications that lead to an emission of light of other wavelengths than green. The origin of GFP dates back to the discovery of a luminescent protein in Aequorea victoria, a jelly fish, by the scientist Osamu Shimomura in the 1960s. Recombinant expression of GFP first succeeded in 1994. The first crystallization of GFP succeeded in 1996 allowing discerning the 3D structure of the protein. The GFP structure consists of different sheets surrounding the chromophore in the center. This insulation of the chromophore allows for a more stable light-reaction of the GFP.
Because GFP expression is not limited to certain tissues GFP can be used in the most various types of model organisms ranging from bacteria to mammals. Today GFP is intensively applied in various labeling applications, such as expression systems of recombinant proteins, localization and tracking of fusion proteins and quantitative measurements of protein content. Mutations of GFP have lead to fluorescent variants of the protein in the colors red, brown, yellow, pink and blue with different intensities. Examples of these multi-colored fluorescent proteins derived from green fluorescent protein are EBFP, Azurite, cyan fluorescent protein (CFP, CyPet, Cerulean etc.), yellow fluorescent protein (YFP, Citrine etc.), red fluorescent protein and several others.
In 2008 the Nobel price for chemistry was awarded for the work on GFP, underscoring the importance that is attributed to the green fluorescent protein in biochemical and molecular biological research.
