Antigen
Alternative Name Annexin V (ANXA5 ELISA Kit Abstract)
Target Type Chemical
Background Annexins are a family of calcium-dependent phospholipid-binding proteins. They are abundant in the eukaryotic kingdom. Though structurally well investigated the in vivo function of the annexins is still unclear. They definitively belong to a family of ubiquitous cytoplasmic proteins involved in signal transduction. All annexins have been shown to have a putative binding site for protein kinases C but only annexin V possesse a potential pseudo-substrate site. Thus annexin V seems to modulate the activity of some PKCs on their substrates. Annexin V was found to play a major role in matrix vesicleinitiated cartilage calcification as a collageregulated calcium channel. Annexin V binds to procoagulant phospholipids (Vascular anticoagulant alpha) with high affinity. Annexin V's preferential binding partner is phosphatidylserine (PS). PS is predominantly located in membrane leaflets, which face the cytosol.

Application Notes Optimal conditions to be determined by end-user
Plate Pre-coated
Assay Procedure An antihuman Annexin V coating antibody is adsorbed onto microwells. Human Annexin V present in the sample orstandard binds to antibodies adsorbed to the microwells. A biotinconjugated antihuman Annexin V antibody is added and binds to human Annexin V captured by the first antibody. Following incubation unbound biotinconjugated antihuman Annexin V antibody is removed during a wash step. StreptavidinHRP is added and binds to the biotinconjugated antihuman Annexin V antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of human Annexin V present in the sample or standard. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A standard curve is prepared from 7 human Annexin V standard dilutions and human Annexin V sample concentration determined.
