Regulated inhibition of programmed cell death (apoptosis) preserves normalhomeostasis and tissue and organ morphogenesis. Aberrations in this processcontribute to human diseases and cancer by abnormally prolonging cell viability. Recently, several apoptosis inhibitors related to the baculovirus iap gene have beenfound in various species, including human. IAP proteins contain one/three Cys/Hisbaculovirus IAP repeats plus a C-terminal RING finger and are thought to block anevolutionary conserved step in apoptosis.Survivin encodes a structurally unique inhibitor of apoptosis (IAP). Survivin expressionis turned off during fetal development and is not found in non-neoplastic adult humantissues. Survivin becomes abundantly re-expressed in transformed cells and in all ofthe most common cancers of lung, colon, pancreas, breast and prostate in vivo. Survivin appears to be situated at the crossroads of cell death and cell division,governing a checkpoint involved in cytokinesis while also suppressing apoptosis. Survivin is also abundantly expressed in brain tissues (astrocytes and some neurons) ofadult rats following traumatic brain injury. Survivin has been found co-expressed withNeuN (mature neuronal marker) and PCNA (a cell cycle protein). Survivin might affectregulation of neural cell proliferative responses after brain injury. This Survivin (60.11) antibody is useful for Flow Cytometry, Immunocytochemistry/Immunofluorescence and Immunoprecipitation. Immunohistochemistry-Paraffin and Western Blot were reported in scientific literature. This antibody weakly detects Survivin in Western blot, for this application we suggest using NB500-235 instead. Use in Immunohistochemistry-Frozen reported in scientific literature (PMID 22841823)

|Protocol||Protocol specific for Survivin Antibody Western Blot Procedure 1) HeLa whole cell lysates were heated to 70 degrees C for 10 minutes and then microfuged at room temperature. 2) 65 mg of lysate were electrophoresed (200 V) through a 4-12 % NuPAGE Bis-Tris gel, for 1 hr. 3) Proteins were transferred (30 V) onto a nitrocellulose membrane for 40 minutes. 4) The membrane was incubated for 1 hr. at ~ 27 degrees C (RT) in blocking buffer [TBST / 5 % nonfat milk]. 5) Rinsed the membrane twice in TBS. 6) Incubated the membrane with a dilution of anti-Survivin antibody, diluted in TBS + 1 % BSA, overnight at 4degrees C, gently shaking. 7) Washed the membrane in TBST, 1x15 min, 3x5min. 8) Incubated the membrane with a goat anti-mouse secondary antibody, diluted in TBS + 1 % BSA, gently shaking for 35 minutes at room temperature. 9) Washed the membrane in TBST, 1x15 min, 3x5min. 10) Visualized membrane by ECL (AlphaInnotech ). NOTE: HeLa whole cell extracts were used as a positive control for this antibody.|
