Specificity	For characterization of different stages of infection and very useful for the analysis of the AAV assembly process. A20 specifically reacts with intact adeno-associated virus particles, empty and full capsids. Recognizes a conformational epitope of assembled capsids, not present in denatured capsid proteins and native but unassembled capsid proteins. The antibody cannot be used for immunoblotting. Epitope mapping experiments (Wobus et al., see below) identified four immunoreactive (discontinous) regions. The major reaction was attributed to sequence aa 369 to aa378 of AAV-2 capsids. The antibody is also useful for neutralizing experiments (cf. Moskalenko et. al). Antibody reacts with AAV-2, found in human and monkey (species tested so far). In ELISA also applicable to AAV-3

Application Notes	Immunofluorescence microscopy ( 1/20, overnight incubation at 2-8°C). Immunohistochemistry on frozen sections ( overnight incubation at 2-8°C) Immunoprecipitation (1/5). Affinity chromatography. ELISA. Neutralization assay. Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user.
Buffer	This antibody is a Protein A affinity purified immunoglobulin fraction and was lyophilised from PBS buffer with 0.09% sodium azide as preservative and 0.5% BSA as stabilizer Reconstitute with 1 ml distilled water. Dilute with PBS or Tris-buffered saline
Storage	Store at 2-8°C. Shelf life: one year after reconstitution.
