| Specificity | Cortactin is an 80/85 kDa cytoskeleton protein that facilitates assembly of cortical actin. Cortactin is widely expressed in most adherent cells and is a prominent substrate of protein-tyrosine kinase Src in vivo and in vitro. The protein sequence of cortactin contains multiple amino-terminal tandem repeats of a unique 37-amino acid sequence, which associates with actin, and a Src homology 3 (SH3) domain at the carboxyl terminus. Between the repeat and the SH3 domain, there is an a-helical structure followed by a proline-rich region. Between the proline-rich region and the SH3 domain, there are a series of Src phosphorylation sites (Tyr-421, Tyr-470, and Tyr-486). In vitro, cortactin binds to and cross-links F-actin into meshworks. Mouse cortactin. Does not cross-react with human. |

| Application Notes | The antibody has been used for Western blot applications, and immunocytochemistry. For Western blot applications, we recommend using the antibody at 0.1-1.0 µg/mL. At 0.35 µg/mL, the dilution provides 100 mL working solution, which at 10 mL/blot allows 10 blots to be performed. Positive control used: 5HD47, a C3H 10T1/2 cell line overexpressing Src and transfected with mouse Cortactin. Other applications not tested. Optimal dilutions of this antibody are dependent on conditions and should be determined by the user. 1 |
