Description	The ion channels activated by glutamate are typically divided into two classes. Thosethat are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors(NMDAR). The NMDAR plays an essential role in memory, neuronal development and ithas also been implicated in several disorders of the central nervous system includingAlzheimer’s, epilepsy and ischemic neuronal cell death. Overexpression of the NR2Bsubunit of the receptor has been associated with increases in learning and memorywhile aged, memory impaired animals have deficiencies in NR2B expression. Tyr1472on NR2B is phosphorylated and this may lead to the increased expression of theNMDAR at the synapse that plays a role in synaptic plasticity.

Application Notes	Antibody dilutions and tissue load should be based on tissue type and expectedphosphorylation state expression levels. Initial recommended range of dilutions: 1:1000. Internally tested under ideal conditions: DB: 1:1000WB: 1:1000 (rat brain extracts)IHC: Not DeterminedNOTE: Immunolabeling is blocked by the phosphopeptide but not by thedephosphopeptide. The antibody also labels bands at ~65kDa and ~115kDa. Human,mouse, and rat have 100% amino acid sequence identity with the antigen used to raisethe antibody.
Buffer	0.1 ml in 10mM HEPES (pH 7.5), 150mM NaCl, 0.1 mg/ml BSA and 50% glycerol.Sequential chromatography on phospho- and non-phosphopeptide affinity columns.Adequate amount of material to conduct 10-mini Western blots.
Storage	For long term storage –80 degrees C is recommended. Short term storage at –20degrees C is acceptable as aliquots are in the presence of 50% glycerol. Stock solutionsare stable for a minimum of 1 year at –20 degrees C.
