Description
|
The beta-adrenergic receptor kinase 1 (betaARK1) was originally isolated as a ser/thr kinase that phosphorylates cardiac beta-adrenergic receptors. It has subsequently been found to phosphorylate many other kinds of G protein coupled receptors (GPCR), as well. It is now recognized as one of a family of G protein coupled receptor kinases that are important in regulating the activity of GPCRs. Phosphorylation of GCPRs by betaARK1, and other related kinases, generally occurs only after the receptor has been activated by ligand binding. The phosphorylation results in the uncoupling of the GPCR from G proteins by binding to beta-arrestin. The binding of beta-arrestin may also result in receptor internalization, mediated by clatherin and dynamin and/or the activation of the MAP kinase cascade. ß,ARK1 is found in many cell types and may also have a role in the regulation of the microtubular cytoskeleton.
