| Description | CD26 is a widely distributed 110 kDa membrane-bound glycoprotein that has a dipeptidyl peptidase IV (DPP IV) activity in its extracellular domain. This molecule is expressed in wide variety of normal tissues and cell populations including lymphocytes. On T cells, its expression is preferentially restricted to the CD4 + helper/memory population, and CD26 can deliver a potent T cell co-stimulatory activation signal. It is also reported that thyroid carcinoma expresses aberrant CD26 activity and that enzyme-histochemical staining of CD26 is very useful for pathological diagnosis of thyroid tumors. |

| Application Notes | Flow cytometric analysis for lymphocytes 1) Preparation of the lymphocytes from whole blood as below. 1. Prepare 5 ml of whole blood pretreated with EDTA. 2. Add 1. onto the 10 ml of Ficoll-paque 3. Centrifuge at 1600 rpm for 30 min. at room temperature. 4. Remove supernatant and collect the Immunohistochemical detection of human CD26 on paraffin embedded section of human Thyroid carcinoma. lymphocytes’ layer by careful pipetting. 5. Add appropriate volume of PBS and mix gently. 6. Centrifuge at 1600 rpm for 8 min. at 4°C. 7. Remove supernatant and mix the cells by gentle tapping, then add appropriate volume of PBS. 8. Centrifuge at 1000 rpm for 8 min. at 4°C. 9. Repeat steps 7. and 8. three times. 2) Prepare the indicated solutions as below. Wash solution: 2% FCS/0.1%NaN3 in PBS 1st antibody solution: anti-Human CD26 (DPP IV) antibody (ready to use) 2nd antibody solution: Anti-Mouse IgG conjugated with FITC (IMMUNOTECH: Cat No. sections: ABC method 1) Deparaffinize section, hydrate to PBS. (Xylene 3 times, Ethanol 3 times, PBS 3 times, immerse the section for 3-5 minutes each.) 2) Remove slides from PBS and cover each section with 100-200 µl of 3% H2O2 for 10 min. at room temperature to block endogenous peroxidase activity. Do not treat frozen section with 3% H2O2. Wash twice in PBS for 5 min. each. 3) Remove slides from PBS, wipe gently around each section and cover tissues with 100-200 µl of Protein Blocking Agent (Ultratech HRP Kit, IMMUNOTECH Cat# IM-2391) for 5min. to block non specific antibody staining. Do not wash. 4) Tip off the blocking buffer, wipe gently around each section and cover tissues with 100-200 µl of primary antibody solution at the concentration of as suggested in Applications. 5) Incubate the section for 1 hr. at room temperature (or 37°C for 10 min.). 6) Wash the slide twice in PBS for 5 min. each. 7) Wipe gently around each section and cover tissues with 100-200 µl of Polyvalent Biotinylated antibody (Ultratech HRP Kit). 8) Incubate for 30 min. at room temperature. 9) Wash as in 6). 10) Wipe gently around each section and cover tissues with 100-200 µl of Streptavidin-Peroxidase (Ultratech HRP Kit) 11) Incubate for 30 min. at room temperature. 12) Wash as in 6). 13) Visualize with DAB substrate/chromogen (Ultratech DAB, IMMUNOTECH Cat# IM-2394) for 5 min. *DAB is a suspected carcinogen and must be handled with care. Always wear gloves. 14) Wash as in 6). 15) Counterstain in hematoxylin for 2min. 16) Wash as in 6). 17) Dehydrate by immersing in Ethanol 3 times for 3 min. each followed by immersing in Xylene twice for 3min. each. 18) Mounting. 050630-1 Immunohistochemistry: ready-to-use.
