| Description | Essential pre-mRNA splicing factor required early in spliceosome formation. Binds to the mammalian polypyrimidine tracts. Forms a complex with the polypyrimidine tract-binding protein (PTB). Seems to also bind DNA. It is a heterotetramer of two 52 kDa and two 100 kDa subunits. The RNAs that direct protein synthesis in animals and plant cells are synthesized in the nucleus as large precursors (pre-mRNAs). The protein coding sequences in pre-mRNA molecules are arranged in discontinuous segments-exons interspersed with noncoding sequences-introns. In a process termed splicing, these introns are efficiently removed before the pre-mRNA is transported from the nucleus to the cytoplasm, where it is translated into protein. Studies have shown that nuclear pre-mRNA splicing takes place in a multi-component structure termed a spliceosome. | Application Notes | For AP: Use at an assay dependent dilution (see Shav-Tal et al reference). For ICC: Use at an assay dependent dilution (fix cells 2 min in 3% paraformaldehyde with 0.5% Triton X-100, and then 20 min in 3% paraformaldehyde) (see Lee et al reference). For IHC-Fr: Use at an assay dependent dilution (4% paraformaldehyde-0.5% Triton X-100 treated tissue). For IP: Use at an assay dependent dilution (see Lee et al reference). For RIA: Use at an assay dependent dilution (see Lee et al reference). WB: Use at a concentration of 2-4 ug/ml. Detects a band of approximately 47, 49, 100, 68, 120, 100, 80 and 68 kDa (predicted molecular weight: 76 kDa). (See Lee et al reference, Shav-Tal et al reference). Not tested in other applications.
