| Application Notes | SDS PAGE & Western Blotting 1) Suspend 1x10 7 of cells with 1 ml of Laemmli sample buffer and sonicate the sample briefly on ice. Make sure the cells are completely lysed. 2) Boil the samples for 2 minutes and centrifuge. Load 10 µ l of a supernatant (cell lysate) per lane in a 1 mm thick 12.5% SDS-polyacrylamide gel for electrophoresis. 3) Blot the protein to a polyvinylidene difluoride (PVDF) membrane at 1 mA/cm 2 for 1 hour in a semi-dry transfer system. (Transfer Buffer - 25 mM Tris, 190 mM glycine, 20% MeOH). See the manufacture's manual for specific transfer procedure. 4) To reduce nonspecific binding, soak the membrane in Block Ace TM buffer for 1 hour at 37°C, or overnight at 4°C. 5) Prepare 1 µ g /ml of anti-HMGB1/2 antibody diluted with 10% Block Ace TM -PBS. Incubate the membrane with the solution for 1 hour at 25°C. (The optimal antibody concentra- ttion will depend on the experimental conditions.) 6) Wash the membrane with 0.2% Tween 20-PBS. (10 minutes x 3 times) 7) Prepare POD-conjugated anti-mouse IgG (MBL, Cat. No. 330) diluted 1:10000 with 10% Block Ace TM -PBS solution as a secondary antibody. Incubate the membrane with the secondary antibody for 1 hour at 25°C. 8) Wash the membrane with 0.2% Tween 20-PBS. (10 minutes x 3 times) 9) Wipe excess buffer from the membrane, then incubate it with appropriate chemiluminescence reagents for 1 minute. Remove extra reagent from the membrane by dabbing with a paper towel, and seal it in plastic wrap. 10) Expose toX-ray film in a dark room for 30 seconds. Develop the film as usual. The conditions for exposure and development may vary. Western blotting: 1 µ g/ml Research Applications. |
