| Description | Interleukin 13 (IL-13) is a T cell derived cytokine involved in the regulation of inflammatory and immune responses. IL-13Ralpha1 together with IL-4Ralpha forms a functional receptor for both IL-4 and IL-13, which is why these two cytokines share many of their biological activities. The receptor is found on human B cells, monocytes and endothelial cells. However, no functional receptor is expressed on T cells, which explains why IL-13, in contrast to IL-4, fails to induce TH2-cell differentiation. Purified GM1E7 was analysed for potential inhibitory effects on IL-13-induced short-term and long-term cellular responses. To analysise potential antagonistic effects of GM1E7 on IL-13-driven cell proliferation, the IL-13-responsive human cell line TF-1 was employed. Cells were treated with IL-13 in the presence of varied concentrations of purified GM1E7. To prove that GM1E7 specifically inhibits the formation of a signalling competent IL-4R alpha/IL-13R alpha1 heterodimer, but does not interfere with the closely related IL-4R alpha/gamma c complex, a parallel experiment was performed with IL-4 as the mitogen. As shown in Fig. 2, antibody GM1E7 blocks IL-13-dependent cell proliferation completely, but only marginally influences IL-4-activity on TF-1 cells. The IC50 for GM1E7 with regard to IL-13-driven proliferation is in the range of 50 nM. |
