The present study was aimed at examining the acrosin activity variations in acrosome exocytosis and its regulation by tyrosine kinase, protein kinase C and voltage-dependent calcium channels.
acrosin was isolated from turkey spermatozoa
During epididymal maturation of boar sperm, alpha-/beta-acrosin isoform expression increases, reaching maximum in cauda; gamma-acrosin isoform is not expressed. In vitro capacitation leads to relocation of acrosin to apical ridge of sperm head.
Data indicate that the positive expression of studied genes fertilization rate for GAPDHS (show GAPDHS Proteins) positive subset was 66%, ACR - 71%, SPATA22 - 68%, MND1 (show MND1 Proteins) - 70%, pregnancy rates were 8%, 6%, 18% and 36% respectively.
Sperm DNA damage is closely related with sperm-nucleoprotein transition, acrosin activity and seminal parameters.
DNA immunization against proacrosin impairs fertility in male mice.
Binding sites for recombinant human zona pellucida (ZP) glycoproteins located at the N- and C-termini of proacrosin reveal a key role of the proenzyme in the interaction.
SERPINA3K (show SERPINA3K Proteins) is involved in the Clock gene-mediated male fertility by regulating acrosin activity and provide the first evidence that SERPINA3K (show SERPINA3K Proteins) could be regulated by Clock gene via retinoic acid-related orphan receptor (show NR1D2 Proteins) response elements.
ACR or PRSS21 (show PRSS21 Proteins) or both may function cooperatively with SPAM1 (show SPAM1 Proteins) in sperm/cumulus penetration
These data demonstrate that the trypsinlike protease activity of ACR and PRSS21 (show PRSS21 Proteins) is essential for the process of sperm penetration through the cumulus matrix and zona pellucida in vitro
Acrosin is the major proteinase present in the acrosome of mature spermatozoa. It is a typical serine proteinase with trypsin-like specificity. It is stored in the acrosome in its precursor form, proacrosin. The active enzyme functions in the lysis of the zona pellucida, thus facilitating penetration of the sperm through the innermost glycoprotein layers of the ovum. The mRNA for proacrosin is synthesized only in the postmeiotic stages of spermatogenesis. In humans proacrosin first appears in the haploid spermatids.
