  • Application Notes
Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing.
    Comment

Tissue Specificity: Produced by kidney or liver of adult mammals and by liver of fetal or neonatal mammals.

    Protocol
rat EPO ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for EPO has been precoated onto 96-well plates. Standards(E.coli, T44-K226) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for EPO is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the rat EPO amount of sample captured in plate.
    Assay Procedure

Aliquot 0.1 mL per well of the 3000pg/mL, 1500pg/mL, 750pg/mL,375.pg/mL, 187.5pg/mL, 93.8pg/mL, 46.9pg/mL rat EPO standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of rat cell culture supernatants, serum, plasma(heparin) or tissue homogenates to each empty well. See "Sample Dilution Guideline" above for details. We recommend that each rat EPO standard solution and each sample is measured in duplicate.

    Background

Protein Function: Erythropoietin is the principal hormone involved in the regulation of erythrocyte differentiation and the maintenance of a physiological level of circulating erythrocyte mass.

Background: Erythropoietin, EPO, also known as hematopoietin or hemopoietin, is a glycoprotein hormone that controls erythropoiesis, or red blood cell production. It is a cytokine for erythrocyte (red blood cell) precursors in the bone marrow. This gene is mapped to 7q22. It is said that the EPO gene encodes a deduced 193-amino acid propolypeptide. This hormone can be found in kidney and liver. It is the hormone that regulates red blood cell production. And it plays an important role in the brain's response to neuronal injury. What’s more, EPO is also involved in the wound healing process.
