Cyclic AMP levels and neurite outgrowth promoted by G protein-coupled receptors

GPR3, GPR6, and GPR12 belong to a family of active G protein-coupled receptors (GPCRs) which share a high similarity at the amino acid level.
The GPCRs are highly expressed in the central nervous system. cAMP production is stimulated by their expression in various cell lines. As Tanaka et al show in their current study that constitutively active GPCRs enhance neurite outgrowth when they are overexpressed in rat cerebellar granule neurons in culture. Thereby GPCRs help to overcome growth inhibition caused by myelin-associated glycoprotein (MAG). The authors conclude that expression of the constitutively active GPCRs upregulates cAMP production in neurons, stimulates neurite outgrowth and counteracts myelin inhibition.
