Plexin B1 and the semaphorin signalling pathways influence the development of prostate cancer in two different ways. Either the elevated expression of Plexin B1 in the majority of primary tumours is crucial or the numerous somatic missense mutations that were detected in the cytoplasmic domain of the Plexin-B1 gene by Oscar Gee-Wan Wong and his team from the University College London (UK) are relevant.

The mutations impede Rac and R-Ras binding and R-RasGAP activity. This results in increased cell motility, invasion, adhesion, and lamellipodia extension.
Semaphorins control cell movement via their transmembrane receptors, the plexins. The scientists figured, that the semaphorin signalling pathway in control of cell migration could be used by cancer cells for invasion and metastasis.
