Molecular dissection of NRG1-ERBB4 signaling implicates PTPRZ1 as a potential schizophrenia susceptibility gene

The genes encoding MAGI and RPTPbeta are significantly associated with schizophrenia. The gene PTPRZ1 that codes for RPTPbeta therefore seems to be involved in the etiology of schizophrenia as are RPTPbeta signalling abnormalities.
The scientists from the Mount Sinai School of Medicine (USA) analyzed data about neuregulin and the neuregulin receptor ERBB4, both being genetically and functionally implicated in schizophrenia to build up a study about finding genes and pathways contributing to schizophrenia susceptibility.
The MAGI scaffolding proteins were discovered to be ERBB4-binding proteins. ERBB4 expression, alone or in combination with ERBB2 or ERBB3, led to the tyrosine phosphorylation of MAGI proteins. The phosphorylation could be enhanced through receptor activation via neuregulin.
MAGI proteins interact with receptor phosphotyrosine phosphatase beta/zeta (RPTPbeta) and RPTPbeta seems to modulate ERBB4 signalling, providing further support for a crucial role of neuregulin/ERBB4 signalling in the molecular basis of schizophrenia.
