|Background||Interleukin 1 receptor, type I (IL1R1), also known as CD121a (Cluster of Differentiation 121a), is an interleukin receptor that belongs to the IL-1 receptor family. IL1R1 is a receptor for both interleukin alpha (IL1A) and interleukin beta (IL1B), and also associates with interleukin-1 receptor antagonist protein (IL-1RA). There are two distinct types of receptors that bind the pleiotropic cytokines IL1A and IL1B: the type I is an 80 kDa transmembrane protein expressed predominantly by T cells, fibroblasts, and endothelial cells and appears to mediate all the known IL-1 biological functions. However, the type II receptor is a 68 kDa transmembrane protein with a short cytoplamic domain and serves as a decoy for IL-1. IL1R1 structurally consists of three C2-type Ig-like domains and one TIR domain. As important mediators involved in many immune and inflammatory responses, the agonists IL1A and IL1B bind to IL1R1, leading to the signal transduction and activation of NF-kappa- B. Formation of a ternary complex containing IL1RAP, TOLLIP, MYD88, and IRAK1 or IRAK2 is indepensible for the signaling processes. Recombinant soluble form of IL1R1 is thus a potent antagonist of IL-1 function.|
