Background TNF- alpha is secreted by macrophages, monocytes, neutrophils, T-cells (principallyCD4+), and NK-cells. Many transformed cell lines also secrete TNF- alpha. Monomerichuman TNF- alpha is a 157 amino acid protein (non-glycosylated) with a reportedmolecular weight of 17 kD. TNF- alpha forms multimeric complexes, stable trimers aremost common in solution. A 26 kD membrane form of TNF- alpha has also beendescribed. TNF- alpha binding to surface receptors elicits a wide array of biologicactivities including: cytolysis and cytostasis of many tumor cell lines in vitro,hemorraghic necrosis of tumors in vivo, increased fibroblast proliferation, and enhancedchemotaxis and phagocytosis in neutrophils. The MAb11 antibody can neutralize thebioactivity of natural or recombinant TNF- alpha. Application Notes The purified MAb11 has been tested by blocking fluorochrome conjugated MAb11 for intracellular cytokine staining. In order to obtain complete blocking results, a saturated amount of purified antibody ( le 5.0 µg/million cells) should be used for incubation with target cells, prior to staining with fluorochrome conjugated antibody. Immunohistochemistry-frozen/paraffin. It is recommended that the reagent be titrated for optimal performance for each application.
