Background Cyclin D1 belongs to the Cyclin D family. Cyclin D1 is required for the cell cycle G1/Stransition (1). Cyclin D1 forms a serine/threonine kinase holoenzyme complex with andfunctions as a regulatory subunit of CDK4 or CDK6. Cyclin D1 also phosphorylates andinactivates the retinoblastoma protein and promotes progression through the G1/Sphase (2-3). Amplification or overexpression of cyclin D1 plays a pivotal role in thedevelopment of various human cancers including parathyroid adenoma, breast cancer,colon cancer, lymphoma, melanoma, and prostate cancer (4-5). Over-expression ofcyclin D1 is known to correlate with the early onset of cancer and risk of tumorprogression and metastasis (6). Cyclin D1 also associates with, and regulates theactivity of transcription factors, coactivators and corepressors that govern histoneacetylation and chromatin remodeling proteins (7-8).
