Human SET Domain and Mariner Transposase Fusion Gene (SETMAR) interaction partners

  1. These results suggest that Metnase enhances Exo1 (show EXO1 Proteins)-mediated exonuclease (show EXO1 Proteins) activity on the lagging strand DNA by facilitating Exo1 (show EXO1 Proteins) loading onto a single strand gap at the stalled replication fork.

  2. The SET domain is needed for the 5' end of ss-overhang cleavage with fork and non-fork DNA without affecting the Metnase-DNA interaction. This domain has a positive role in restart of replication fork and the 5' end of ss-overhang cleavage.

  3. methylation of snRNP70 (show SNRNP70 Proteins) by SETMAR regulates constitutive and/or alternative splicing

  4. Metnase may possess an important role in DNA repair, topoisomerase II (show TOP2 Proteins) function, and the maintenance of stemness during colon cancer development.

  5. 293 T transfected with Metnase revealed a large number of rescued plasmids.

  6. found known and novel SETMAR splice variants to be significantly increased in acute myeloid leukemia (show BCL11A Proteins)

  7. a single mutation DDN(610) --> DDD (show KRT5 Proteins)(610), which restores the ancestral catalytic site, results in loss of function in Metnase

  8. phosphorylation of Metnase S495 differentiates between these two functions, enhancing DSB repair and repressing replication fork restart.

  9. Data show that DBN1 (show DBN1 Proteins), SETMAR and HIG2 (show HILPDA Proteins) are direct transcriptional targets of the SOX11 (show SOX11 Proteins) protein.

  10. DNA repair protein (show ATRIP Proteins) Metnase (also SETMAR), which has a SET histone methylase domain, localized to an induced DSB and directly mediated the formation of H3K36me2 near the induced DSB

Mouse (Murine) SET Domain and Mariner Transposase Fusion Gene (SETMAR) interaction partners

  1. The suppression of Notch-1 (show NOTCH1 Proteins) led to increased expression of Oct4 (show POU5F1 Proteins) and Sox2 (show SOX2 Proteins), but in decreased gene expression of cMET, Setmar and CD44 (show CD44 Proteins).

  2. Metnase may have been selected for because it has a function opposing transposases and may thus play a key role in suppressing translocations that underlie oncogenicity.

SETMAR Protein Profile

Protein Summary

This gene encodes a fusion protein that contains an N-terminal histone-lysine N-methyltransferase domain and a C-terminal mariner transposase domain. The encoded protein binds DNA and functions in DNA repair activities including non-homologous end joining and double strand break repair. The SET domain portion of this protein specifically methylates histone H3 lysines 4 and 36. This gene exists as a fusion gene only in anthropoid primates, other organisms lack mariner transposase domain. Alternate splicing results in multiple transcript variants.
