    Characteristics
Plate 3*104 cell per well in 96-well plates in RPMI 1640 medium with 10%FBS for 24 h. Sub confluent cells were starved 4 h in RPMI 1640 medium with 2%FBS. Standard and Sample 2-human TNF- alpha were added and cell viability measured 24 h later by addition of CellTiter 96® AQueous Non-Radioactive Cell Proliferation Assay reagent (MTS). The ED50 for this effect is typically 0.02-0.08ng/ml.

    Background
TNF-alpha plays a major role in growth regulation, differentiation, inflammation, viral replication, tumorigenesis, and autoimmune diseases. Besides inducing hemorrhagic necrosis of tumors, TNF has been found to be involved in tumorigenesis, tumor metastasis, viral replication, septic shock, fever, inflammation, and autoimmune diseases including Crohn's disease, and rheumatoid arthritis as well as graft-versus-host disease. TNF alpha-1a is a potent lymphoid factor that exerts cytotoxic effects on a wide range of tumor cells and certain other target cells. Synonym: TNF-alpha. Formulation: Lyophilized from a 0.2 mym filtered solution in PBS.
