There are two kinds of IGF receptors based on their relative affinities for IGF1 versusIGF2. The IGF1 receptor prefers IGF1 over IGF2 and weakly binds insulin. IGF1receptor is a disulfide-linked heterotetrameric transmembrane protein consisting of twoalpha (130 kD) and two beta (95 kD) subunits. Both the alpha and beta subunits areencoded within a single receptor precursor cDNA. The IGF1 receptor is therefore similarin structure to the insulin receptor. The proreceptor polypeptide is proteolytically cleavedand disulfide-linked to yield the mature heterotetrameric receptor. The IGF1 receptor ishighly expressed in all cell types and tissues and is highly overexpressed in mostmalignant tissues where it functions as an anti-apoptotic agent by enhancing cellsurvival. Protocol specific for IGF1 Receptor Antibody Deparaffinization:
1. Incubate slide at 60C for 60 minutes.
. Deparaffinize in Xylene for 10 minutes and repeat one more times.
. Hydrate in 100 % alcohol for 5 minutes, in 95 % alcohol for 5 minutes, in 85 % alcohol for 5 minutes, in 75 % alcohol for 5 minutes.
. Dip into Distill Water for 5 minutes.
. Dip into TBS (50 mM Tris, 100 mM NaCl, pH 7.6), leave for 5 minutes, and repeat two times.Antigen Retrieval:
. Bring 500 - 2000 mL 10 mM citrate buffer (pH6.
0) to the boil in a stainless steel pressure cooker.
. Put the slide into staining rack and lower into pressure cooker ensuring the slide is well immersed in citrate buffer.
. When the pressure indicator valve has risen after 3 to 4 minutes, incubate for 1 minute.
. Cool the slide naturally to room temperature.
. Dip into distilled water, leave for 5 minutes, and repeat two times.
. Dip the slide in TBS for 5 minutes and repeat two times.
. Immerse slides in 3 % H2O2 ( in fresh methanol) for 15 minutes at room temperature.
. Wash with distilled water two times, 5 minutes each time.
. Wash with TBS (pH 7.
) two times, 5 minutes each time.Staining with Primary Antibody:
. Dilute primary antibody with 3 % BSA in TBS. Cover the tissue section on the slide with diluted primary antibody (use 50 to 150ul for each slide).
. Incubate at 37 degrees C for 30 minutes or at room temperature for 60 minutes (The optimal incubation time, incubation temperature, and antibody dilution should be determined by the individual laboratory).
. Wash with TBS two times, 5 minutes each time.Staining with Secondary Antibody:
. Incubate with 100-200ul Polymer Enhancer. Incubate 30 minutes at 37 degrees C.
. Wash with TBS for 3 times, 5 minutes each time.
. Incubate with 100-200ul Polymerized HRP and incubate 30 minutes at 37 degrees C
. Wash with TBS for 3 times, 5 minutes each time.
