|Background||The platelet-derived growth-factor (PDGF) family induces a variety of cellular responses including cell proliferation, migration, differentiation, and matrix deposition. The PDGF ligands are dimeric molecules encoded by three different genes, PDGF-A, -B, -C, and the recently described PDGF-D. Two tyrosine kinase receptors, PDGFR-alpha and PDGFR-beta, are also known CD140a and CD140b respectively, become activated upon ligand binding and dimerization. The binding of effector proteins to the activated PDGFR results in the activation of multiple signal transduction pathways. PDGFR-alpha binds PDGF-A, -B and -C, whereas PDGFR-beta binds PDGF-B and -D. The pleiotropic effect of PDGFs on various cell types and the expression of PDGFR-alpha in a variety of tissues throughout embryonic development and postnatal life suggest its roles in multiple processes such as gastrulation, vasculogenesis, wound healing, and glial cell development.Synonyms: Alpha-type platelet-derived growth factor receptor, CD140 antigen-like family member A, PDGF Receptor alpha, PDGF-R-alpha|

Flow Cytometry: 5-10 μg/mL (final concentration). Positive Control: Human Transfectant. Detailed procedure is provided in Protocols.

Optimal dilutions are dependent on conditions and should be determined by the user.
|Protocol||Flow cytometric analysis for adherent cellsWe usually use Fisher tubes or equivalents as reaction tubes for all step described below. 1) Detach the cells from culture dish by trypsinization. *Excessive trypsinization mayreduce the antigenecity. 2) Wash the cells 3 times with washing buffer [PBS containing 2% fetal calf serum (FCS)and 0. 1% NaN]. 3) Resuspend the cells with washing buffer (5x106 cells/mL)4) Add 50 µL of the cell suspension into each tube, and centrifuge at 500 x g for 1 minute atRT (20~25°C). Remove supernatant by careful aspiration. 5) Add 10 µL of normal goat serum containing 1 mg/mL normal human IgG and 0. 1% NaN3to the cell pellet after tapping. Mix well and incubate for 5 minutes at RT. 6) Add 40 µL of the FITC labeled anti-PDGF receptor alpha/CD140a antibody (Clone 16A1)(12. 5-25 µg/mL) diluted with the washing buffer. Mix well and incubate for 30 minutes atRT. 7) Add 1 mL of the washing buffer followed by centrifugation at 500 x g for 1 minute at RT. Remove supernatant by careful aspiration. 8) Resuspend the cells with 500 µL of the washing buffer and analyze by a flow cytometer. Positive Control: Human Transfectant.|
