Vascular endothelial growth factor (VEGF) was recently identified as a secreted,direct-acting mitogen specific for vascular endothelial cells and capable of stimulatingangiogenesis in vivo. Molecular cloning revealed multiple forms of VEGF, apparentlyarising from alternative splicing of its RNA transcript (1). VEGF and TGF-beta1 arepotent angiogenesis inducers with opposing effects on endothelial cells. TGF-beta1induces apoptosis, VEGF protects endothelial cells from apoptosis. We found thatTGF-beta1 promotes endothelial cell expression of FGF-2, which up-regulates VEGFsynthesis. Inhibition of VEGF signaling through VEGF receptor 2 (flk-1) abrogatesTGF-beta1-induced apoptosis and p38(MAPK) activation. This finding illustrates anunexpected role of VEGF and indicates that VEGF can be pharmacologically convertedinto an apoptotic factor, a novel approach to antiangiogenesis therapy (2).
