Background In humans, CD4 is expressed on the surface of peripheral T-cells, thymocytes, on macrophages and granulocytes. It acts as a cellular adhesion molecule binding MHC class 2 molecules. Moreover, CD4 also acts as a receptor for HIV but requires cosignals. CD4 expression in brain suggests that it may play a more general role in mediating cell recognition events than merely those of cellular immune responses. CD4(+) cells are T-helper cells which, upon cell activation, produce a number of different cytokines and play a role in the activation and/or proliferation of B-cells, cytotoxic T-lymphocytes, and macrophages. Thymocytes which are CD4(+) and CD8(-) produce factors such as IL4, IL5, IL10 and IFN-gamma. CD4 is a 55 kDa transmembrane glycoprotein belonging to the immunoglobulin superfamily.

Application Notes Blocking, staining and activating antibody. Blocks blinding of HIV to CD4. For staining, use 10 µl/10⁶ cells. Titer for blocking and activating T cells should be determined individually by the investigator. Antibody might also be suited for other applications not tested so far.

Handling Advice Please Note: Always centrifuge product briefly before opening vial. Reconstitute with 0.5 ml sterile H20 or PBS to give a 1 mg/ml concentration. Mix gently, wash the sides of the vial and wait 30-60 seconds before use. Protein concentration: 1 mg/ml (after reconstitution).
