These results demonstrate that Ascl3 marks progenitors that are lineage-committed strictly to microvillar cells and Bowman's glands, and highlight the requirement for these cell types to support OE homeostasis.
Ascl3 knockouts develop smaller salivary glands than wild types, but secrete saliva (show RAG1AP1 Proteins) normally. They display a lower level of cell proliferation, consistent with their smaller size. In the absence of Ascl3, the cells maintain their progenitor function.
These data demonstrate a progenitor-progeny relationship between duct cells and the acinar cell compartment, and identify a population of multipotent progenitor cells, marked by expression of Ascl3, which is capable of generating both gland cell types.
