These data provide a genetic basis for the unstable phenotype of Helios-deficient Tregs within the inflammatory environment of tumors and suggest that immune milieu-dependent alterations in gene expression are a central feature of Treg conversion.
Helios controls certain aspects of Treg-suppressive function, differentiation, and survival.
this study reports that impaired expression of the transcription factor Helios by FoxP3 (show FOXP3 Antibodies)+ CD4 (show CD4 Antibodies) and Qa-1 (show H2-T23 Antibodies)-restricted CD8 (show CD8A Antibodies) Tregs results in defective regulatory activity and autoimmunity in mice.
data suggest that a subset of DN T cells, identified by the expression of PD-1 (show PDCD1 Antibodies) and Helios, represent self-reactive cells; results provide an explanation for the origin of DN T cells and introduce CD8 (show CD8A Antibodies) loss as a process associated with self-Ag encounter
results demonstrate that Helios is expressed by thymic and peripheral T cells that are being driven to tolerance in response to a genuine autoantigen
identify Helios as the first marker known to diverge during positive and negative selection of thymocytes
Helios regulates IL-2 (show IL2 Antibodies) production in Tregs by suppressing Il2 (show IL2 Antibodies) gene transcription. Loss of Helios in Tregs breaks their anergic phenotype and results in derepression of the Il2 (show IL2 Antibodies) locus.
Helios expression spatially coincides with the early pattern of expression of the mature markers of striatal projection neurons.
In tumor-bearing mice, Helios(+) regulatory T cells were relatively over-represented in the tumor-mass.
Data found that Helios-negative T cells are enriched for naive T cell phenotypes and vice versa. Moreover, Helios can be induced during T cell activation and proliferation, but regresses in the same cells under resting conditions.
mRNA expression of Helios was decreased in CD4 (show CD4 Antibodies)+ T cells from patients with acute coronary syndrome.
High expression of TIGIT (show TIGIT Antibodies) and Helios identifies CD4 (show CD4 Antibodies)+ T cells with impaired immunological functions, primarily among patients with an advanced stage of Sezary syndrome.
revealed more than 170 NFAT (show NFATC1 Antibodies)-associated proteins, half of which are involved in transcriptional regulation. Among them are many hitherto unknown interaction partners of NFATc1 (show NFATC1 Antibodies) and NFATc2 (show NFAT1 Antibodies) in T cells, such as Raptor (show RPTOR Antibodies), CHEK1 (show CHEK1 Antibodies), CREB1 (show CREB1 Antibodies), RUNX1 (show RUNX1 Antibodies), SATB1 (show SATB1 Antibodies), Ikaros (show IKZF1 Antibodies), and Helios.
this study shows that Ikaros (show IKZF1 Antibodies) undergoes a transient increase in protein levels at the transitional single-positive CD8 (show CD8A Antibodies)+ developmental stage before diverging in their expression patterns at later stages
The results show that high vs. low expression of Nrp-1 (show NELL1 Antibodies) or Helios does not unequivocally identify Treg clones of thymic or peripheral origin.
Helios could be considered a more reliable marker for distinguishing thymic derived Regulatory T (tTreg) cells or peripherally induced Treg cells than Nrp1 (show NELL1 Antibodies).
Forkhead box (show FOXO Antibodies) protein 3 (show HSPB3 Antibodies)(+) regulatory T cells and Helios(+) subset in perinatally acquired HIV.
The immunosuppressive enzyme IL4I1 (show IL4I1 Antibodies) expressed differentially in human induced Aiolos (show IKZF3 Antibodies)+, but not natural Helios+, FOXP3 (show FOXP3 Antibodies)+ Treg cells.
Ectopic expression of ATL-type Helios isoform as well as knockdown of normal Helios or Ikaros (show IKZF1 Antibodies) promoted T-cell growth.
Foxp3 (show FOXP3 Antibodies)+ Helios+ regulatory T cells are expanded in active systemic lupus erythematosus.
This gene encodes a member of the Ikaros family of zinc-finger proteins. Three members of this protein family (Ikaros, Aiolos and Helios) are hematopoietic-specific transcription factors involved in the regulation of lymphocyte development. This protein forms homo- or hetero-dimers with other Ikaros family members, and is thought to function predominantly in early hematopoietic development. Multiple transcript variants encoding different isoforms have been found for this gene, but the biological validity of some variants has not been determined.
