    Peptide ELISA: BLNK antibody Limit Dilution: 1/32000. Western blot: 1-3 μg/mL. Approx 70 kDa band observed in lysates of cell line Daudi(calculated MW of 50.0 kDa according to 037446.1). The observed molecular weightcorresponds to how SLP-65 is expected to run. However, when transiently expressed inHEK293, a 50 kDa band is observed.

    Background
    BLNK (B cell linker protein) is an adaptor molecule that plays key role in B cell activation initiated by cross-linking the B cell receptor (BCR). This activation leads to an increase in the tyrosine phosphorylation of a number of proteins due to activation of nonreceptor protein tyrosine kinases (PTK) including Syk, which is responsible for the tyrosine phosphorylation of BLNK. BLNK is cytosolic protein identified as two phosphoproteins migrating at 68 and 70 kDa in SDS/PAGE (alternatively spliced forms of human BLNK). Mapping of the tyrosine residues in BLNK reveal three tyrosines that bind PLC-gamma2, and two tyrosines that bind the Tec family kinase Btk, the Vav nucleotide exchange factor and the adaptor Nck.Synonyms: B-cell adapter containing a SH2 domain protein, B-cell adapter containing a Src homology 2 domain protein, BASH, Cytoplasmic adapter protein, SLP-65, SLP65, Src homology 2 domain-containing leukocyte protein of 65 kDa
