  • Application Notes
    Peptide ELISA: Limit dilution 1: 32000. Western blot: Recommended for use at 1-2 μg/mL. Approx 70 kDa and 50 kDa bands seen inJurkat cell lysate. Note the smaller isoform of this protein has a consensus glycosylationsite, which may explain the higher than expected band size (50 kDa versus 30 kDa). Glycosylation can affect migration of proteins on SDS-PAGE [Hames, B. D and Rickwood, D. ,eds. Gel electrophoresis of proteins: a practical approach. Oxford University Press].
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Background
    ELMO1 is involved in cytoskeletal rearrangements required for phagocytosis of apoptotic cells and cell motility. ELMO1 acts in association with DOCK1 and CRK and was initially proposed to be required in complex with DOCK1 to activate Rac Rho small GTPases. It is belived to enhance the guanine nucleotide exchange factor (GEF) activity of DOCK1. ELMO1 interacts directly with the SH3-domain of DOCK1 via its SH3-binding site. It is part of a complex with DOCK1 and RAC1 and with DOCK1 and CRK isoform CRK-II.
