    mouse IL15RA ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. Monoclonal antibody from rat specific for IL15RA has been precoated onto 96-well plates. Standards(NSO, G33-K205) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for IL15RA is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse IL15RA amount of sample captured in plate.
    Assay Procedure

    Aliquot 0.1 mL per well of the 8000pg/mL, 4000pg/mL, 2000pg/mL, 1000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL mouse IL15RA standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of mouse cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each mouse IL15RA standard solution and each sample be measured in duplicate.

    Background: Interleukin 15 receptor, alpha subunit is a subunit of the interleukin 15 receptor which is encoded by the gene IL15RA in humans. The human IL15RA gene was mapped to chromosome 10p15-p14 by FISH and the mouse Il15ra gene was mapped to chromosome 2. The cytoplasmic domain of IL15R-alpha, like that of IL2R-alpha, was dispensable for mitogenic signaling, suggesting that the primary role of the alpha chains is to confer high-affinity binding. At high concentrations, IL15, like IL2, was able to signal through a complex of IL2R-beta and -gamma in the absence of the alpha subunit. The soluble receptor displayed high affinity for IL15 and inhibited both binding of IL15 to the membrane receptor and IL15-induced cell proliferation.
