    This COVID19 human IgG antibody ELISA kit employs an indirect ELISA method. In this kit, standard 96-well plates (12 strips with 8 wells/strip) are coated with the SARS-CoV-2 N protein, which combines with the corresponding antibody present in a sample and Positive Control which used as calibration curve for normalization purpose. The wells are washed and biotinylated anti-human IgG antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of COVID19 N protein human IgG antibody bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    Positive Control is from inactivated serum sample which contains SARS-COV-2 N protein human IgG antibody. We do not know the exact amounts of SARS-COV-2 N protein human IgG antibody in the Positive Control sample. The Positive Control only can be used as calibration curve for normalization purpose in different assays.
    Assay Procedure
    1. Prepare all reagents, samples and standards as instructed.
    2. Add 100 μL positive control or sample to each well. Incubate 1.5 hours at room temperature.
    3. Add 100 μL prepared HRP- anti positive control IgG Antibody and/or 100 μL of prepared HRP-anti Human IgG Antibody to each well. Incubate 1 hour at room temperature.
    4. Add 100 μL TMB One-Step Substrate Reagent to each well. Incubate 10 minutes at room temperature.
    5. Add 50 μL Stop Solution to each well. Read at 450 nm immediately.
    6. Read at 450 nm immediately.
    Calculation of Results

    Calibration curve: Calculate the mean absorbance for each set of duplicate Positive Control (Item C), samples, and subtract the average zero Positive Control optical density. Plot the calibration curve on log-log with Positive Control concentration (Unit/mL) on the xaxis and absorbance on the y-axis using Sigma plot or Excel software.
