Protocol
  • Intracardially perfuse the animal with the fixative solution (4% (w/v) PFA, 15% v/v picric acid saturated solution). Remove the brain and postfixate in the same solution ON at RT.
  • Cut slices (70µm of thickness) in a vibratome.
  • Proceed with a free-floating IHC in a 24-well plate (1 slice, 250µl of volume per well).
  • Wash slices for 10min with 0.1M PBS pH7.4 at RT.
  • Permeabilize slices 3x for 5min with 0.1M PBS containing 1% v/v triton x-100 (PBST).
  • Block non-specific binding with staining solution (PBST containing 10% v/v fetal bovine serum) for 1h at RT.
  • Incubate slices with primary rabbit anti-Chromogranin A (CHGA) antibody (antibodies-online, ABIN617895, lot 1436001) diluted 1:1000 in staining solution ON at RT.
  • Wash slices 3x for 5min with PBST.
  • Incubate slices with secondary goat anti-rabbit IgG, biotin-conjugated antibody (Vector, BA-1000, lot W1002) diluted 1:200 in staining solution for 2h at RT.
  • Wash slices 3x for 5min with PBS.
  • Incubate slices with ABC kit (Vectastain, PK-6100) diluted 1:100 in staining solution for 1h at RT.
  • Wash slices 3x for 5min with PBS.
  • Incubate with 0.05% DAB (Sigma, D5637) and 0.01% H2O2 in PBS, watching reaction at the microscope.
  • Wash slices 3x for 5min with PBS.
  • Mount slices on gelatinated glass slides, dehydrate and coverslip with DPX mountant (VWR, 360294H).
  • Image acquisition with Leica SP-5 confocal microscope, 10x and 20x, resolution 1368x1024.
Experimental Notes
  • In macaque sections the labeling obtained with ABIN617895 was the same as previously described in human hippocampus (Munoz (1990)).

  • In mouse hippocampus, however, staining was only observed in unexpected scattered cells, probably interneurons. In rat hippocampus, it was not detected any clear labeling. Further tests are needed to determined if the antibody is not properly detecting murine CHGA or if the antigen is not present in the hippocampus in these two species.
