    Brand
    BD Pharmingen™
    Specificity
Biological Activity: The activity range is measured in an anti-viral assay using A549 indicator cells
Specific Activity: 0.1 - 2.0 x 10^8 Units/mg (Unit is defined as the amount of material required to stimulate a half-maximal response at cytokine saturation.)
    ED50: 0.05 - 1.0 ng/ml
Observed dose response relationship: 16 fold Since applications vary, each investigator should titrate the reagent to obtain optimal results.
    Characteristics
Recombinant human IFN-gamma is supplied as a frozen liquid comprised of 0.22 µm sterile-filtered aqueous buffered solution containing 5.0 mg/ml Biotechnology grade, low endotoxin bovine serum albumin, with no azide. Human IFN-gamma was found to be ≥ 95% pure by SDS-PAGE analysis and an absorbance assay based on the Beers-Lambert law.

  • Application Notes
ELISA Standard: Recombinant human IFN-gamma is useful as a quantitative standard for measuring human IFN-gamma protein levels in a sandwich ELISA with purified NIB42 (Cat. No. 551221) as a capture antibody and the biotinylated 4S.B3 (Cat. No. 554550) as the detection antibody.
Note: This ELISA pair is recommended primarily for measuring cytokine from experimental cell culture systems. These ELISA reagents are not recommended for assaying serum or plasma samples. For measuring IFN-gamma in serum or plasma our BD OptEIA™ Human IFN-gamma Set (Cat. No. 555142 ) or OptEIA™ Human IFN-gamma Kit (Cat. No. 550612 ) are specially formulated and recommended.

    Handling Advice
Interferon-γ (IFN-γ) is a potent multifunctional cytokine which is secreted by activated NK cells and CD4+TCRαβ+, CD8+TCRαβ+, and TCRγδ+ T cells. IFN-γ exerts its biological effects through specific binding to a single class of high affinity receptors. In addition to its antiviral effects, IFN-γ can upregulate a number of lymphoid cell functions including the antimicrobial and antitumor responses of macrophages, NK cells, and neutrophils. In addition, IFN-γ can exert strong regulatory influences on the proliferation, differentiation, and effector responses of B cell and T cell subsets. These influences can involve IFN-γ's capacity to boost MHC class I and II expression by antigen-presenting cells as well as to direct effects on B cells and T cells themselves. Human IFN-γ is a 14 - 18 kD multiple glycosylated protein containing 143 amino acid residues.
