Characteristics: The recombinant mouseCD14 is produced from mouse CD14 transfected CHO-cells in serum free medium, has been purified by His-tag with Talon (Clontech) and controlled by SDS page. Before transfection the complete mouse CD14-cDNA was amplified by PCR and cloned into expression vector p-POL-DHFR.

Storage/Stability: Long time storage at -70oC. When stored at recommended temperature, this protein is stable for 12 months.The lyophilized product thought stable at -70oC more than 1 year, at refrigerator about 10 months at room temperature up to 4 weeks, and at 37oC 1 week. Reconstituted protein should be used immediately or stored in undiluted working aliquots at -20oC. For long term storage it is recommended to add a carrier protein (o.1% BSA). Repeated freezing and thawing should be avoided.

Background: Background: CD14 acts as a receptor for endotoxin (LPS), expressed strongly on monocytes, macro-phages and weekly at surface of neutrophiles. CD14 is anchored to cells by linkage to glycosylphosphatidylinositol (GPI) and functions as a high affinity receptor LPS-LBP (lipopolysaccharide binding protein)-complexes. CD14 cooperates with MD-2 and TLR4 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Up-regulates cell surface molecules, including adhesion molecules.CD14 is expressed strongly on the surface of monocytes and weakly on the surface of granulocytes, also expressed by most tissue macrophages.
