|Application Notes||Western Blot 1:1000, Chromatin Immunoprecipitation 1 μg antibody/ 10 μg protein, Flow Cytometry 8 μg/mL where cells are used at 2*10^6/mL, ELISA 1:100-1:2000, Immunohistochemistry 1:200, Immunocytochemistry/Immunofluorescence 1:50-1:200, Immunoprecipitation, Immunohistochemistry-Paraffin 1:200, Immunohistochemistry-FrozenThis Blimp-1 (3H2-E8) antibody is useful for ELISA, Immunocytochemistry/Immunofluorescence, Western Blot, Immunohistochemistry on paraffin-embedded sections, Flow Cytometry, Immunoprecipitation and Chromatin Immunoprecipitation applications. Immunohistochemistry-Frozen was reported in scientific literature. By WB, this antibody recognizes a band at ~98 kDa and may recognize one at ~80 kDa (the beta form). Antigen retrieval is recommended (EDTA buffer, microwave) prior to IHC on paraffin tissues. This antibody demonstrates nuclear staining. For IHC and ICC/IF a dilution of 1:200 is recommended with tyramide amplification.|
The antibodies are intended for use in vitro experiments only. Our antibodies have not been tested nor are recommended for use in vivo.
Protocol specific for Blimp-1 Antibody Western Blot
1. Run ~20 micrograms of protein on a 10 % SDS-PAGE.
. Block nitrocellulose membrane with 5 % blotto in TBS-0.2 % Tween-20, overnight at 4 degrees Celcius.
. Incubate membrane with diluted anti-Blimp-1 (primary antibody, cat# NB 600-235) for 1 hour at RT, in 2 % blotto in TBS-0.2 % Tween-20.
. Wash blot 4 times for 10 minutes, each, at RT with TBS-0.2 % Tween-20.
. Incubate membrane with anti-mouse IgG1 conjugated to HRP (secondary antibody), diluted in 2 % blotto in TBS-0.2 % Tween-20, for 45 minutes at RT.
. Wash 4 times for 10 minutes, each, at RT with TBS-0.2 % Tween-20.
. Incubate membrane with chemiluminescence reagent as per vendor specification. Dry blot and cover with saran wrap.
. Expose to film.9.Develop image.Immunohistochemistry / ImmunofluorescencePlease refer to the following refernence:Cattoretti, G, et al. PRDM1/Blimp-1 is expressed in human B-lymphocytes committed to the plasma cell lineage.J Pathol. 206(1):76-86 (2005)
