Pyruvate is an intermediate in the network of metabolic pathways. Pyruvate can be converted into carbohydrates via gluconeogenesis, to fatty acids or energy through acetyl-CoA (citric acid cycle), to the amino acid alanine, and to ethanol. Therefore, it unites several key metabolic processes.

Pyruvic acid supplies energy to cells through the citric acid cycle when oxygen is present (aerobic respiration), and alternatively ferments to produce lactate when oxygen is lacking (fermentation).

With their Pyruvate Colorimetric & Fluorometric Assay Kit (ABIN411703) BioVision provides a simple and automated procedure to measure pyruvate in samples such as blood, cells or culture media. In the assay, pyruvate is oxidized by pyruvate oxidase via enzyme reactions to generate color (λ= 570 nm) and fluorescence (at Ex/Em = 535/587 nm). It is able to detect 1 μM to 10 mM pyruvate.

Trypsin Activity Colorimetric Assay Kit (ABIN411710)

Trypsin is a serine protease, found in the digestive system of many vertebrates. It catalyzes the hydrolysis of peptide bonds, breaking down proteins into smaller peptides. Trypsin cleaves peptide chains mainly at the carboxyl side of the amino acids lysine or arginine, except when either is followed by proline. The cleavage occurs within the polypeptide chain rather than at the terminal amino acids, therefore trypsin is considered as an endopeptidase. The process is commonly referred to as trypsin proteolysis or trypsinisation.

Trypsin is available in high quantity in pancreases, and can be purified rather easily. Hence it has been used widely in various biotechnological processes. Trypsin can be utilized to resolve cells adherent to the cell culture dish wall prior to harvesting. In proteomics it is the go-to enzyme to digest proteins into peptides for mass spectrometry analysis, e.g. in-gel digestion. The high specificity, due to hydrolization restrictions mentioned above, argue for trypsin usage.

The assay cleaves a substrate to generate p -nitroaniline (p -NA) which is detected at λ =450nm. Glutathione (GSH) can be found in almost all cells in high concentration and works as an important antioxidant in plants, animals, fungi, and some bacteria and archaea. The low-molecular-weight thiol plays a critical role in the cellular defense against oxidative stress such as free radicals, peroxides, lipid peroxides, and heavy metals in mammalian cells. Glutathione reduces disulfide bonds formed within cytoplasmic proteins to cysteines by serving as an electron donor. In the process, glutathione is converted to its oxidized form, glutathione disulfide (GSSG). Glutathione reductase is able to reduce back glutathione to its original state, using NADPH as an electron donor. The ratio of reduced glutathione to oxidized glutathione within cells is often used as a measure of cellular oxidative stress.

The ApoGSH™ Glutathione Colorimetric Assay Kit (ABIN411672) provides a sensitive, colorimetric method for detecting the GSH changes in apoptosis and other pathological processes. Both reduced and oxidized forms can be analyzed from tissues, cells, plasma & erythrocytes. The assay is based on the glutathione recycling system by DTNB and glutathione reductase. DTNB and GSH react to generate 2-nitro-5-thiobenzoic acid which has yellow color. In order to determine the GSH concentration measure absorbance at 412 nm. The generated GSSG can be reduced back to GSH by glutathione reductase, and the reaction repeats, forming more 2-nitro-5-thiobenzoic acid. Therefore, the recycling system dramatically improves the sensitivity of total glutathione detection. The kit can quantify glutathione from 1-100 ng/well in a 200 l reaction. With increase of reaction time even lower glutathione concentrations, such as in blood samples can be detected. Alkaline Phosphatase Activity Colorimetric Assay Kit (ABIN411784)

Alkaline phosphatase (AP) is a hydrolase enzyme responsible for removing phosphate groups from many types of molecules, including nucleotides, proteins, and alkaloids. The process of removing the phosphate group is called dephosphorylation. As the name suggests the optimal working environment for alkaline phosphatases is slightly basic. Changes in alkaline phosphatase level and activity are associated with various disease states in the liver and bone.

By changing the amino acids of the wild-type alkaline phosphatase enzyme produced by E.coli, a mutant alkaline phosphatase was created. It is 36 times more active compared to the enzyme acitivity of standard alkaline phosphatase while remaining stable. Alkaline phosphatase has become a useful tool in molecular biology laboratories. DNA normally possesses phosphate groups on the 5' end. Removing these phosphates prevents the DNA from ligating, which is undesirable during plasmid DNA cloning; also, removal of the phosphate groups allows radiolabeling in enzyme immunoassays. The alkaline phosphatase from shrimps is easy to inactivate with heat and therefore easy to regulate in an experiment.
