|Background||FGFR4, also designated as CD334, is a member of the fibroblast growth factor receptor subfamily which belongs to the tyrosine protein kinase superfamily. Upon ligand binding, FGFRs undergo dimerization and autophosphorylation, intiating a cascade of downstream signal pathways, ultimately influencing a variety of physiological and pathological cellular processes including mitogenesis and differentiation. FGFR members is highly conserved in amino acid sequence and differ f in their ligand affinities and tissue distribution. FGFR4, a type I transmembrane glycoprotein, contains 3 immunoglobulin-like domains in the extracellular domain and a cytoplasmic tyrosine-kinase domain. FGFR4 expression is detected in gastrointestinal epithelial cells, pancreas, as well as gastric and pancreatic cancer cell lines. FGFR4 is reported to be a high affinity receptor for both acidic and basic fibroblast growth factor, and also binds to FGF19. Like FGF R1- 3,several isoforms of FGFR4 are generated by alternative splicing of the mRNAs. Although its specific function is unknown, growing studies have indicated that FGFR4 plays a role in tumorigenesis and malignace of cetain carcinomas such as breast and ovarian tumors.|
