Usage: Blocking of receptor-ligand interaction-Approximately 4 to 10 μg/mL of this antibody will block 50 % of the binding of 25 ng/mL of biotinylated rhAng-2 to immobilized rzfTie-2/Fc chimera (100 μL of a 4 μg/mL solution was coated in each well) in a functional ELISA. Approximately 50 μg/mL of this antibody will block 95 % of the binding. Western blot-This antibody can be used at 0.1-0.2 μg/mL with the appropriate secondary reagents to detect zebrafish Tie-2. The detection limit for rzfTie-2 is approximately 2 ng/lane under non-reducing and reducing conditions. Immunohistochemistry-This antibody has been used at a concentration of 10 μg/mL to detect Tie2 in 24hpf zebrafish embryos. Dechorionated zebrafish embryos were fixed overnight in 4 % paraformaldehyde at 4° C and then dehydrated and rehydrated through a series of methanol washes. Embryos were blocked in PBS containing 0.1 % Tween 20, 2 % normal donkey serum, and 1 % BSA for 2 hours at room temperature. After blocking, embryos were incubated in diluted primary antibody overnight at 4 °C followed by. donkey anti-goat IgG at room temperature for 2 hours. Between each step, embryos were washed in PBS containing 0.1 % Tween 20. Direct ELISA-This antibody can be used at 0.5-1.0 μg/mL with the appropriate secondary reagents to detect zebrafish Tie-2. The detection limit for rzfTie-2 is approximately. 0.1 ng/well.
