Background Programmed cell death 4 (Pdcd4) is a novel tumor suppressor. Pdcd4 directly inhibits thehelicase activity of eukaryotic translation initiation factor 4A (eIF4A), a component of thetranslation initiation complex. Pdcd4 also suppresses the transactivation of activatorprotein-1 (AP-1)- responsive promoters by c-Jun. Pdcd4 contains two Akt phosphorylationsites, one at Ser67 and the other at Ser457. The phosphorylation of Pdcd4 by Akt causesnuclear translocation of Pdcd4 and a significant decrease in the ability of Pdcd4 tointerfere with the transactivation of AP-1-responsive promoters by c-Jun.

Application Notes ELISA: 1: 400,000. Western blot: 1: 1,000 - 1: 3,000. Immunohistochemistry on frozen sections: 1.25 - 2.5 µg/ml, we recommend the use of freshfrozen tissues. Attempts at staining paraffin-embedded formalin fixed tissues were negative. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Storage Comment Store the antibody at 2 °C to 8 °C up to one month or (in aliquots) at -20 °C to -70 °C forlonger. To minimize loss of volume (25µl) dilute 1: 10 by adding 225 µl of the buffer statedabove directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect thevolume at the bottom of the vial. Use this intermediate dilution when calculating finaldilutions. Avoid repeated freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. Shelf life: one year from despatch.
