Importins, Impbeta, Kapbeta2, Imp4 (show SPPL2B Antibodies), Imp5 (show IPO5 Antibodies), Imp7 (show IPO7 Antibodies), Imp9 (show IPO9 Antibodies), and Impalpha, show the H3 tail binding more tightly than the H4 tail. The H3 tail binds Kapbeta2 and Imp5 (show IPO5 Antibodies) with KD values of 77 and 57 nm, respectively, and binds the other five Importins more weakly.
Study identified a core region encompassing nt - 118 to + 108 of IPO4 gene that is necessary for its promoter activity. Transcription factors binding to this region were screened, resulting in the identification of two members of the Ets (show ETS1 Antibodies) family, Ets (show ETS1 Antibodies)-like transcription factor-1 (show HNF1A Antibodies) and GA binding protein, which repress or activate its promoter activity, respectively.
report a function of IPO4 and nuclear import in the Fanconi anemia (show PALB2 Antibodies) pathway of DNA repair
demonstrated that vitamin D receptor (VDR) has two nuclear import systems: ligand-dependent & ligand-independent pathways, & that importin 4 fulfills the ligand-independent nuclear transport through the interaction with the amino terminus of VDR (show CYP27B1 Antibodies)
importins 4 and 7 accomplish nuclear import of HIF-1alpha (show HIF1A Antibodies) more efficiently than the classical importin alpha (show KPNA4 Antibodies)/beta NLS (show ALDH1A2 Antibodies) receptor.
Metallothionein (show MT Antibodies) deficient mice have a marked decrease in Importin 4 transcripts during acute lung injury
Functions in nuclear protein import as nuclear transport receptor. Serves as receptor for nuclear localization signals (NLS) in cargo substrates. Is thought to mediate docking of the importin/substrate complex to the nuclear pore complex (NPC) through binding to nucleoporin and the complex is subsequently translocated through the pore by an energy requiring, Ran- dependent mechanism. At the nucleoplasmic side of the NPC, Ran binds to the importin, the importin/substrate complex dissociates and importin is re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran. The directionality of nuclear import is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus (By similarity). Mediates the nuclear import of RPS3A. In vitro, mediates the nuclear import of human cytomegalovirus UL84 by recognizing a non-classical NLS.
