Mouse (Murine) Purine-Rich Element Binding Protein B (PURB) interaction partners

  1. site-directed mutagenesis revealed several basic amino acid residues in the intermolecular (R267) and intramolecular (K82 and R159) subdomains that are essential for Purbeta transcriptional repressor function in Acta2 (show ACTA2 ELISA Kits) promoter-reporter assays. In keeping with their diminished Acta2 (show ACTA2 ELISA Kits) repressor activity in fibroblasts, purified Purbeta variants containing an R267A mutation exhibited reduced binding affinity for purine-ric (show FXYD5 ELISA Kits)...

  2. This structural arrangement suggests that the cooperative assembly of the dimeric Purbeta repressor on the sense strand of the ACTA2 (show ACTA2 ELISA Kits) enhancer is dictated by the association of each subdomain with distinct purine-rich binding sites within the enhancer.

  3. These findings suggest that the highly conserved modular repeats of Purbeta fold to form a core functional domain, which mediates the specific and high affinity binding of the protein to single-stranded DNA.

  4. Overexpression of Purbeta, but not Puralpha, significantly downregulated GnRH1 (show GNRH1 ELISA Kits) mRNA levels in transiently transfected GT1-7 cells, suggesting that Purbeta acts as a repressor of GnRH1 (show GNRH1 ELISA Kits) gene transcription.

  5. Purbeta is a single-stranded DNA-binding repressor of vascular smooth muscle alpha-actin (show ACTA2 ELISA Kits) gene transcription

  6. analysis of of DNA binding and localized strand separation by Pur alpha and comparison with Pur beta

  7. Pur alpha and Pur beta repress smooth muscle alpha-actin gene transcription by means of DNA strand-selective cis-element binding and cell type-dependent protein-protein interactions

  8. analysis of the quaternary structure of recombinant mouse Purbeta

  9. These data demonstrate that Pur proteins collaborate with Sp3 to regulate a transcriptional program that enables muscle cells to remodel their phenotype.

  10. structural role for certain core arginine residues in forming a conformationally stable Pur beta protein capable of physical interactions necessary for smooth muscle alpha-actin (show ACTA2 ELISA Kits) gene repression.

Human Purine-Rich Element Binding Protein B (PURB) interaction partners

  1. Results showed that Purbeta ssDNA-binding activity was significantly increased in myeloid cells from AML (show RUNX1 ELISA Kits) patients suggesting that the enhanced ssDNA-binding activity of Purbeta in transformed myeloid cells may serve as a unique and measurable phenotypic trait for improving prognostic risk stratification in AML (show RUNX1 ELISA Kits).

  2. site-directed mutagenesis revealed several basic amino acid residues in the intermolecular (R267) and intramolecular (K82 and R159) subdomains that are essential for Purbeta transcriptional repressor function in Acta2 (show ACTA2 ELISA Kits) promoter-reporter assays. In keeping with their diminished Acta2 (show ACTA2 ELISA Kits) repressor activity in fibroblasts, purified Purbeta variants containing an R267A mutation exhibited reduced binding affinity for purine-ric (show FXYD5 ELISA Kits)...

  3. Suggest dynamic interplay between transcriptional activators Pur-alpha/Pur-beta and repressors in regulating SMalphaA (show ACTA2 ELISA Kits) gene output during myofibroblast differentiation.

  4. results show that PUR proteins are capable of binding to alpha-MHC (show MYH6 ELISA Kits) mRNA and attenuate its translational efficiency; also show robust expression of PUR proteins in failing hearts where alpha-MHC (show MYH6 ELISA Kits) mRNA levels are suppressed

  5. findings offer insights into mechanism, energetics, and sequence of Purbeta repressor binding to a biologically relevant, contractile phenotype-regulating cis-element while revealing the thermodynamic confines of Purbeta-mediated effects on DNA structure

Purine-Rich Element Binding Protein B (PURB) Antigen Profile

Antigen Summary

This gene product is a sequence-specific, single-stranded DNA-binding protein. It binds preferentially to the single strand of the purine-rich element termed PUR, which is present at origins of replication and in gene flanking regions in a variety of eukaryotes from yeasts through humans. Thus, it is implicated in the control of both DNA replication and transcription. Deletion of this gene has been associated with myelodysplastic syndrome and acute myelogenous leukemia.
