Human Ribosomal Protein L5 (RPL5) interaction partners

  1. Patients with RPS19 mutations had the fewest number of defects, while patients with RPL5 had the greatest number of birth defects. This is the first study to show differences between Diamond-Blackfan anemia (DBA) genetic groups with regards to treatment.

  2. RPL5 remains an interesting candidate in multiple myelom(MM )because it is deleted in 20-40% of MM cases

  3. Whole exome sequencing in the differential diagnosis of Diamond-Blackfan anemia: Clinical and molecular study of three patients with novel RPL5 and mosaic RPS19 mutations

  4. Low RPL5 expression is associated with cancer.

  5. Ribosomal proteins L11 and L5 activate TAp73 by overcoming MDM2 inhibition.

  6. RPL5 mutation is associated with Diamond Blackfan Anemia.

  7. Findings uncover a mechanism by which RPL5 and RPL11 can co-operatively suppress c-Myc expression, allowing a tightly controlled ribosome biogenesis in cells.

  8. Unlike other tumor suppressors, RPL5 and RPL11 play essential roles in normal cell proliferation.

  9. High frequency of RPL5 gene deletion is associated with Italian Diamond-Blackfan anemia.

  10. Oncogenic splicing factor SRSF1 stabilizes the tumor suppressor protein p53 via RPL5, inducing cell senescence.

  11. Mutations affect the ribosomal proteins RPL5 and RPL10 in 12 of 122 (9.8%) pediatric T-cell acute lymphoblastic leukemias.

  12. disrupted nucleoli may provide a platform for L5- and L11-dependent p53 activation, implying a role for the nucleolus in p53 activation by ribosomal biogenesis stress

  13. Data show 1 proband with an RPL5 deletion, 1 patient with an RPL35A deletion, 3 with RPS17 deletions, and 1 with an RPS19 deletion.

  14. Data show that all patients with RPS19 and RPL5 mutations had physical abnormalities.

  15. An analysis and fine mapping of GFI-EVI5-RPL5-FAM69A locus, genotyping eight Tag-single nucleotide polymorphisms in 732 multiple sclerosis patients and 974 controls from Spain, was performed.

  16. Knockdown of L29 or L30 enhanced the interaction of MDM2 with L11 and L5 and markedly inhibited MDM2-mediated p53 ubiquitination, suggesting that direct perturbation of 60 S ribosomal biogenesis activates p53 via L11- and L5-mediated MDM2 suppression.

  17. The study reports a high frequency of RPL5 (9.3%) and RPL11 (9.3%) mutations in a Diamond-Blackfan anemia cohort.

  18. the presence of multiple NLSs in ribosomal protein L5 appears to allow for efficient nuclear transport via utilisation of multiple, mechanistically different import pathways.

  19. the MDM2-L5-L11-L23 complex functions to inhibit MDM2-mediated p53 ubiquitination and thus activates p53

  20. interaction of NVL2 with L5 is ATP-dependent and likely contributes to the nucleolar translocation of NVL2

Mouse (Murine) Ribosomal Protein L5 (RPL5) interaction partners

  1. there is a fine tuned balance in the interaction of ribosomal proteins with the MDM2/p53 axis which is important in normal hematopoiesis.

  2. Ribosomal proteins L11 and L5 activate TAp73 by overcoming MDM2 inhibition.

  3. There was a significant delay in the G2/M phase in Rpl5 mutant cells, unaffected by p53 knockdown.

  1. These results therefore provide a comprehensive basis for the study of molecular pathogenesis of RPL5-mediated Diamond-Blackfan anemia and other ribosomopathies.

RPL5 Protein Profile

Protein Summary

Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal protein that is a component of the 60S subunit. The protein belongs to the L18P family of ribosomal proteins. It is located in the cytoplasm. The protein binds 5S rRNA to form a stable complex called the 5S ribonucleoprotein particle (RNP), which is necessary for the transport of nonribosome-associated cytoplasmic 5S rRNA to the nucleolus for assembly into ribosomes. The protein interacts specifically with the beta subunit of casein kinase II. Variable expression of this gene in colorectal cancers compared to adjacent normal tissues has been observed, although no correlation between the level of expression and the severity of the disease has been found. This gene is co-transcribed with the small nucleolar RNA gene U21, which is located in its fifth intron. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome.
