Human Polyclonal FANCI Primary Antibody for IP, WB - ABIN253065
Colnaghi, Jones, Cotto-Rios, Schindler, Hanenberg, Huang: Patient-derived C-terminal mutation of FANCI causes protein mislocalization and reveals putative EDGE motif function in DNA repair. in Blood 2011
Data suggest that FANCI and FANCD2 (show FANCD2 Antibodies) have partially non-overlapping and possibly even opposing roles during the replication stress response.
Fanconi anemia (show PALB2 Antibodies) FANCD2 (show FANCD2 Antibodies) and FANCI proteins regulate the nuclear dynamics of splicing factors, such as SF3B1 (show SF3B2 Antibodies).
FANCB (show BRCA2 Antibodies) dimer coordinates FANCD2 (show FANCD2 Antibodies):FANCI monoubiquitination by two FANCL (show FANCL Antibodies) RING-ligases. Deubiquitination of FANCD2 (show FANCD2 Antibodies):FANCI by USP1 (show USP1 Antibodies):UAF1 (show WDR48 Antibodies) occurs only when DNA is removed.
depletion of FANCI, but not FANCD2 (show FANCD2 Antibodies) or USP1 (show USP1 Antibodies), results in increased phosphorylation and activation of Akt (show AKT1 Antibodies).
FANCI mutations are associated with Fanconi anemia (show PALB2 Antibodies) in VACTERL association.
FANCJ (show BRIP1 Antibodies) protein is important for the stability of FANCD2 (show FANCD2 Antibodies)/FANCI proteins and protects them from proteasome and caspase-3 (show CASP3 Antibodies) dependent degradation.
These findings indicate that FANCI functions upstream of FA core complex recruitment independently of FANCD2 (show FANCD2 Antibodies), and alter the current view of the FA-BRCA pathway.
these purification methods for human FANCI and FANCD2 (show FANCD2 Antibodies) provide novel procedures to facilitate structural and biochemical studies of human FANCI and FANCD2 (show FANCD2 Antibodies).
Results show that ATR (show ANTXR1 Antibodies)-mediated phosphorylation of FANCI, controls dormant origin firing in response to DNA replication stress.
Our studies reveal a previously unknown mechanism for the coordinate nuclear import of a subset of FANCD2 and FANCI, a key early step in the cellular ICL response.
crystal structure of FANCI-FANCD2 (show FANCD2 Antibodies)(ID) complex; crystallographic electron-density map of FANCI protein bound to splayed Y DNA; data suggest ID complex recognizes DNA structures resulting from encounter of replication forks with an interstrand cross-link
The Fanconi anemia complementation group (FANC) currently includes FANCA, FANCB, FANCC, FANCD1 (also called BRCA2), FANCD2, FANCE, FANCF, FANCG, FANCI, FANCJ (also called BRIP1), FANCL, FANCM and FANCN (also called PALB2). The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity\; they are related by their assembly into a common nuclear protein complex. This gene encodes the protein for complementation group I. Alternative splicing results in two transcript variants encoding different isoforms.
