Human Pleiomorphic Adenoma Gene 1 (PLAG1) interaction partners

  1. PLAG1 is a microRNA target gene that is overexpressed in Wilms tumors with mutations in microRNA processing genes.

  2. HRAS (show HRAS Proteins) mutations were more common in epithelial-myoepithelial carcinomas (EMCAs) with intact PLAG1 and HMGA2. Most EMCAs arose ex pleomorphic adenoma (PA)and the genetic profile of EMCA varies with the absence or presence of preexisting PA and its cytogenetic signature. Progression to higher grade EMCA with intact PLAG1 and HMGA2 correlates with the presence of TP53 (show TP53 Proteins), FBXW7 (show FBXW7 Proteins) mutations, or SMARCB1 (show SMARCB1 Proteins) deletion.

  3. Plag1 expression is lost in malignant transformation from pleomorphic adenoma to carcinoma ex pleomorphic adenoma.

  4. MiR (show MLXIP Proteins)-26a can facilitate occurrence of pituitary tumor and invasiveness, probably via inhibiting PLAG1 expression.

  5. expression of PLAG1 can be considered a valuable diagnostic marker for recurrent pleomorphic adenoma

  6. High PLAG1 expression is associated with pleomorphic adenoma.

  7. The sensitivity (55%) and specificity (75%) of PLAG1 in diagnosing PA in FNAs is relatively modest thus limiting its diagnostic utility.

  8. The Thr34 of PLAG1 is critical for LpMab-10 recognition.

  9. Data indicate that SLIT2 (show SLIT3 Proteins), miR (show MLXIP Proteins)-218-1, RET (show RET Proteins)/PLAG1 and SLIT2 (show SLIT3 Proteins)/ROBO1 (show ROBO1 Proteins) pathway involved in Hirschsprung's disease.

  10. Altogether, these data suggest that HMGA2 is an upstream activator of PLAG1.

PLAG1 Protein Profile

Protein Summary

Pleomorphic adenoma gene 1 encodes a zinc finger protein with 2 putative nuclear localization signals. PLAG1, which is developmentally regulated, has been shown to be consistently rearranged in pleomorphic adenomas of the salivary glands. PLAG1 is activated by the reciprocal chromosomal translocations involving 8q12 in a subset of salivary gland pleomorphic adenomas. Three transcript variants encoding two different isoforms have been found for this gene.
