    Yeast (Saccharomyces cerevisiae)
    Cross-Reactivity (Details)
The antibody does not cross react to any other cellular protein.
    Characteristics
Inp2p- selective antibodies were generated against a peptide taken from the region corresponding to the aa sequence (SVQ GLS LER KVD FID VAE). The Inp2p-selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage. The anti-Inp2p-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions. The Supplier provides Inp2p Western blot positive control samples in SDS-PAGE sample buffer.

    Affinity Purified
    Immunogen
Synthetic peptide corresponding to unique amino acid sequences on Saccharomyces cerevisiae S288c Inp2p protein.
    Isotype
    IgG
  • Application Notes
Antibodies were tested in ELISA and western blotting applications at 1:500 dilution using ABIN1040759 samples. Antibody dilutions for these antibodies are for reference only, investigators are expected to determine the optimal conditions. Application of this antibody in other protocols has not yet tested.
    WB: > 1:500
    IMM & IP pull-down assays: N.D. .
    IHC: N.D.
Investigators using this antibody in protocols other than listed above can request a complimentary sample of this antibody. N.D. not necessarily means the antibody is not suitable for that application, it simply means we have not yet characterized the antibody for that application.

    Background
Inp2p is a peroxisomal receptor protein present in S.cerevisiae that helps in the portioning of the peroxisomes during bud formation. Inp2p accumulates in peroxisomal membrane at the time of budding and interacts with the globular tail of Myo2p (Myosin). Peroxisomes that contain Inp2p are selected and moved to the bud. Inp2p is a phosphoprotein whose level of phosphorylation is coupled to and oscillates with the cell cycle. Inp2p levels on individual peroxisomes are controlled by peroxisome inheritance, as it aberrantly accumulates and decorates all peroxisomes in mother cells when peroxisome partitioning is abolished. Cells overproducing Inp2p often transfer their entire populations of peroxisomes to buds. Cells ramp up Inp2p expression during mitosis, but the receptor only appears on a subset of peroxisomes, potentially selecting them for Myo2p-mediated movement to the bud. Cells lacking Inp2p fail to partition peroxisomes to the bud but are unaffected in the inheritance of other organelles. The gene for Inp2p is present in chromosome XIII in S.cerevisiae. Inp2p have two coiled-coil domains that are each about 30 AA residues in length. Tandem coiled-coil domains of about the same size represent a common feature of all organelle receptors for class V myosins. The levels of Inp2p and those of their corresponding mRNAs, oscillate during the cell cycle in a pattern that parallels the dynamics of yeast organelles, strongly suggesting that the amount of class V myosin receptor associated with an organelle at each point of the cell cycle is crucial to the regulation of the movement of that organelle. A correlation exists between the levels of Inp2p on different peroxisomes and their segregation fates, as only peroxisomes containing detectable amounts of Inp2p are selectively carried to the daughter cell .
