- human IL1R2 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for IL1R2 has been precoated onto 96-well plates. Standards(sf21, F14-E343) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for IL1R2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human IL1R2 amount of sample captured in plate.
- Assay Procedure

Aliquot 0.1 mL per well of the 2000pg/mL,1000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, 62.5pg/mL, 31.2pg/mL human IL1R2 standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of human cell culture supernates or serum to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each human IL1R2 standard solution and each sample be measured in duplicate.

Protein Function: Non-signaling receptor for IL1A, IL1B and IL1RN. Reduces IL1B activities. Serves as a decoy receptor by competetive binding to IL1B and preventing its binding to IL1R1. Also modulates cellular response through non-signaling association with IL1RAP after binding to IL1B. IL1R2 (membrane and secreted forms) preferentially binds IL1B and poorly IL1A and IL1RN. The secreted IL1R2 recruits secreted IL1RAP with high affinity, this complex formation may be the dominant mechanism for neutralization of IL1B by secreted/soluble receptors. .
Background: Interleukin 1 receptor, type II(IL1R2) also known as CD121b(Cluster of Differentiation 121b) is an interleukin receptor. The protein encoded by this gene is a cytokine receptor that belongs to the interleukin-1 receptor family. It is mapped to 2q11.2. IL1R2 binds interleukin alpha(IL1A), interleukin beta(IL1B), and interleukin 1 receptor, type I(IL1R1/IL1RA), and acts as a decoy receptor that inhibits the activity of its ligands. IL1R2 also modulates cellular response through non-signaling association with IL1RAP after binding to IL1B.
