Alternative Name CCL28 (CCL28 ELISA Kit Abstract)
Background

Protein Function: Chemotactic for resting CD4, CD8 T-cells and eosinophils (By similarity). Binds to CCR10 and induces calcium mobilization in a dose-dependent manner. .

Background: CCL28, also known as mucosae-associated epithelial chemokine(MEC), CCK1 and SCYA28, is a chemokine. It is mapped to 5p12. The tumor hypoxia promotes the recruitment of regulatory T(Treg) cells through induction of expression of the chemokine CCL28, which in turn promotes tumor tolerance and angiogenesis. CCL28 regulates the chemotaxis of cells that express the chemokine receptors CCR3 and CCR10. CCL28 has also been implicated in the migration of IgA-expressing cells to the mammary gland, salivary gland, intestine and other mucosal tissues. It has also been shown as a potential antimicrobial agent effective against certain pathogens, such as Gram negative and Gram positive bacteria and the fungus Candida albicans.

Synonyms: C-C motif chemokine 28,Small-inducible cytokine A28,Ccl28,Scya28,

Full Gene Name: C-C motif chemokine 28

Cellular Localisation: Secreted.
Gene ID 56838
UniProt Q9JIL2
Application Notes Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing.
Comment

Tissue Specificity: Mainly expressed in testis, epithelial cells of normal colon, kidney, Peyer patches, lymph nodes. Also found in lower levels in brain, spleen and lung.

Plate Pre-coated
Protocol mouse CCL28 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for CCL28 has been precoated onto 96-well plates. Standards(E.coli, I23-R130) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CCL28 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse CCL28 amount of sample captured in plate.
Assay Procedure

Aliquot 0.1 mL per well of the 2000pg/mL,1000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, 62.5pg/mL, 31.2pg/mL mouse CCL28 standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of mouse cell culture supernates, serum or plasma(heparin, EDTA) to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each mouse CCL28 standard solution and each sample be measured in duplicate.
