Hypothalamic pro-opiomelanocortin and AgRP (show AGRP Antibodies) neurons are hypophysiotropic, projecting to the pituitary to coordinately regulate multiple pituitary hormones.
cloning and characterization of the pro-opiomelanocortin gene
Zebrafish proopiomelanocortin (zfPOMC) contains the consensus sequences for ACTH, gamma-LPH, beta-MSH and beta-endorphin (beta-END). RT-PCR expression studies indicate that zfPOMC is selectively expressed in nervous tissue and in the pituitary gland.
Pituitary expression of proopiomelanocortin provides a unique marker for pituitary anterior and intermediate lobe morphogenesis.

Data suggest that the solitary tract pro-opiomelanocortin (NTSPOMC) neurons were involved in cardiorespiratory control and endogenous analgesia.
hypothalamic arcuate nucleus (Arc (show NOL3 Antibodies))POMC-deficient mice, which develop severe obesity and insulin (show INS Antibodies) resistance, unexpectedly exhibit improved glucose tolerance and remain protected from hyperglycemia via decreased renal GLUT2 (show SLC2A2 Antibodies) expression and glycosuria.
Hypomethylation of POMC promoter occurred in the hypothalamus of the high sucrose diet offspring compared with controls.
Together, these results suggest that alpha-MSH alleviates Dex-induced damages to cultured osteoblasts through activating MC4R (show MC4R Antibodies)-SphK1 (show SPHK1 Antibodies) signaling.
These data indicate that glucose induces distinct excitatory synaptic plasticity in different subpopulations of POMC neurons.
Although genetic deletion of either Atg12 (show ATG12 Antibodies) or Atg5 (show ATG5 Antibodies) renders POMC neurons autophagy-deficient, mice lacking Atg5 (show ATG5 Antibodies) in POMC neurons do not exhibit these phenotypes.
Data indicate that LIM-homeodomain (show LHX2 Antibodies) transcription factor Islet 1 (show ISL1 Antibodies) is necessary for hypothalamic proopiomelanocortin gene (Pomc) expression during embryogenesis.
these results reveal that cooperative interactions between the enhancers confer robustness of Pomc expression against gene regulatory disturbances and preclude deleterious metabolic phenotypes caused by Pomc deficiency in adulthood
In contrast to mice with total hypothalamic Pomc deficiency, which are severely obese, mice with Lepr (show LEPR Antibodies)-restricted Pomc expression displayed fully normal body weight, food consumption, glucose homeostasis, and locomotor activity.
these common variants in PCSK1 (show PCSK1 Antibodies) and POMC were not the major cause of obesity in the Thai subjects sampled. However, variants in PCSK1 (show PCSK1 Antibodies) did affect cholesterol level, LDL-C level, and waist circumference.
ACTH was readily detected at significantly higher levels in regions of ACTH-secreting adenomas and in normal anterior adenohypophysis compared with non-secreting adenoma and neurohypophysis.
CSF (show CSF2 Antibodies) POMC was higher in lean vs. overweight/obese subjects.
There was a non-linear association between leukocyte telomere length and plasma ACTH after laboratory stressor in elderly adults.
Synovial fluid alpha-MSH levels showed an independent and negative correlation with articular cartilage degeneration in patients with knee ACL (show ACLY Antibodies) deficiency.

This gene encodes a polypeptide hormone precursor that undergoes extensive, tissue-specific, post-translational processing via cleavage by subtilisin-like enzymes known as prohormone convertases. There are eight potential cleavage sites within the polypeptide precursor and, depending on tissue type and the available convertases, processing may yield as many as ten biologically active peptides involved in diverse cellular functions. The encoded protein is synthesized mainly in corticotroph cells of the anterior pituitary where four cleavage sites are used\; adrenocorticotrophin, essential for normal steroidogenesis and the maintenance of normal adrenal weight, and lipotropin beta are the major end products. In other tissues, including the hypothalamus, placenta, and epithelium, all cleavage sites may be used, giving rise to peptides with roles in pain and energy homeostasis, melanocyte stimulation, and immune modulation. These include several distinct melanotropins, lipotropins, and endorphins that are contained within the adrenocorticotrophin and beta-lipotropin peptides. Mutations in this gene have been associated with early onset obesity, adrenal insufficiency, and red hair pigmentation. Alternatively spliced transcript variants encoding the same protein have been described.
